Lentiviral vector analytics

Connect vector production and gene-modified cell products to measurements that answer the right biological question.

Conceptual cutaway illustration of a lentiviral vector with an envelope, surface proteins and internal RNA.

The analytical question

Define what the result means.

A lentiviral vector preparation and a population of transduced cells require different analytical evidence. We help define the measurand, sample, reporting unit and decision before selecting a method. The resulting strategy connects molecular measurements with functional studies and the wider product control strategy.

  • RNA genome titerA measurement of assay-detectable vector RNA, commonly reported as RNA copies per volume. Extraction, reverse transcription, DNA contamination and target accessibility can affect the result.
  • Functional or transducing titerA cell-based readout of productive transduction under specified assay conditions. Cell susceptibility, endpoint and study design influence the reported result; RNA copies and particle markers are not substitutes.
  • Cellular vector copy number (VCN)A cellular DNA measurement normalized to a defined denominator. Average copies per total cell and copies per transduced cell are different. Evidence must address nonintegrated DNA before a result is described as integrated VCN.

Potential workstreams

How we can help.

The final scope is agreed for the product, development stage and scientific decision.

Attribute and unit mapping

Separate genome titer, particle-associated measurements, functional activity and cellular VCN across the relevant process stages.

Assay architecture and controls

Review target placement, RNA/DNA specificity, extraction recovery, reverse transcription, inhibition and reference-material suitability.

Cell-product characterization

Plan VCN normalization and, where relevant, integration-site profiling with explicit sampling, detection and interpretation limits.

Impurity and safety strategy

Map residual nucleic acids, process impurities and replication-competent lentivirus testing to distinct, appropriately scoped work packages.

Stability and comparability

Select complementary readouts to investigate change over time or after a process change, without assuming that titer alone captures product quality.

Technical data review

Assess assay assumptions, conflicting readouts and evidence gaps; prioritize additional experiments with the executing laboratory.

Consulting deliverables

What you receive.

Depending on the agreed engagement, outputs can include:

  • A measurand, sample and reporting-unit map
  • An assay and control matrix with documented rationale
  • A VCN interpretation and normalization review
  • A prioritized method-development or troubleshooting plan
  • A brief for separately commissioned laboratory studies

These are proposed advisory outputs, not claims of completed experiments or validated performance. Laboratory execution and provider responsibilities are scoped separately.

Scientific boundaries

Detection of a vector sequence by PCR does not, by itself, demonstrate integration, functional transduction or the absence of replication-competent virus. Integration-site profiling and replication-competent lentivirus testing are distinct applications requiring their own evidence.

Common questions

Is VCN the same as vector titer?

No. VCN describes vector copies in a defined cell population. Vector-stock titer describes a property of the vector preparation, such as assay-detectable genomes or transducing activity per volume.

Can a molecular assay replace functional characterization?

A molecular assay can provide an informative component of a control strategy. It does not automatically replace a product-relevant functional assay or a separately justified potency strategy.
